A High Throughput Method for Generating Dihaploids from Tetraploid Potato

Abstract There is a worldwide effort to increase the efficiency of potato ( Solanum tuberosum L.) cultivar development by using inbred diploid breeding lines. This activity impeded cost and required produce dihaploids from cultivated tetraploid potatoes. We developed high throughput dihaploid production method based on 60-year-old Peloquin Hougas. Red Norland inflorescences commercial fields were transferred greenhouses. As buds developed, pollen inducer IVP 101 was applied systematically thousands stigmas per trial. Berries harvested 21 days after pollination. Seeds putative lacking seed spot marker retained ploidy confirmed flow cytometry. recovered 23 21,651 pollinations. promising for carrying out pollinations since field-grown flowers dramatically less than that greenhouse-grown flowers.
